Identification of F1 Hybrid Progenies in Mango Based on Fluorescent SSR Markers

Mango (Mangifera indica L.) belongs to the genus Mangifera and family Anacardiaceae, is an important tropical fruits. Artificial cross breeding (hand pollination) method for new mango cultivars. It easy produce false hybrids in process of artificial pollination breeding. Therefore, it necessary establish rapid accurate molecular detection methods identify authenticity hybrids. ‘Jinhuang’ ‘Renong No.1′ 65 individual plants their F1 were used as experimental materials, eight SSRs (simple sequence repeats) primer pairs with polymorphism parents by capillary electrophoresis. The results showed that PCR product size (bp) primers ranged from 108 bp (ES55) 176 (ES63) samples. A total 62 true identified hybrid progenies, rate was 95.38%. 18 alleles amplified SSRs, seven SSR loci binary segregations, whereas only one locus ES83 showing ab:ac:bb:bc segregation fitted expected ratio 1:1:1:1. value heterozygosity (He), 0.34 0.62, observed 0.44 0.81. Chi-square test accordance Mendel’s law. cluster analysis could be classified into two categories at 0.58: first category contained 27 offspring, clustered a maternal genetic tendency. second 35 partial paternal DNA fingerprint × constructed using variety protection.
